Abstract The incidence of Salmonella in Egypt was estimated using ISO 6579 (2002) which has detection limit of 100 cfu/25 g and 100% relative accuracy and was compared with polymerase chain reaction. A polymerase chain reaction assay was developed for the generic detection of Salmonella spp. and the identification of S. Enteritidis, S. Gallinarum, S. Pullorum and S. Typhimurium from poultry related samples. Specificity of the assay was tested using 7 Salmonella standard strains and 3 non Salmonella standard strains and 13 isolated Salmonella serovars. The assay was 100% specific for Salmonella detection. At the genus level, peR-RV detected 2 more positive field samples than the standard microbiological techniques and results were obtained faster. A multiplex polymerase chain reaction assay was used for simultaneous detection of S. Enteritidis, S. Gallinarum, S. Pullorum and S. Typhimurium from poultry related samples. |
