Abstract The present study aimed to evaluate the ameliorative effect of Hypericum Perforatum extract on memory dysfunction induced by scopolamine in adult male albino rats (Rattus norvegicus). The study used Memantine HCl as a reference drug approved by FDA for Alzheimer’s disease treatment. Adult male albino rats were treated with daily oral administration Hypericum Perforatum (350 mg/kg) or Memantine Hel (20 mg/kg) for two, four, eight weeks and at the end of each interval the rats were intraperitoneally administered with scopolamine (0.4 mg/kg). All rats were subj ected to Morris water maze (MWM) before and after treatment. Scopolamine administration significantly increased the latency time in Morris water maze at the three durations. Also, it caused significant increase in AChE activity and aspartic acid, glutamic acid, serine, glycine and taurine levels. On the other hand, Scopolamine caused significant decrease in the content of asparagine, GABA, noradrenaline, dopamine, serotonin in the cortex and hippocampus and significant decreasesin the levels of serum T3, T4 and TSH. Administration of Hypericum Perforatum and memantine pre-scopolamine administration caused a decrease in the latency time in MWM and a decrease in the activity of ACHE and the contents of aspartic acid, glutamic acid, taurine, glycine and serine. Also, both Hypericum Perforatum and memantine caused a significant increases in the content of asparagine, GABA, noradrenaline, dopamine, serotonin of the cortex and hippocampus, and the levels of serum T3, T4 and TSH. The study demonstrates a significant neuroprotective effect of Hypericum Perforatum extract which mimics the effect of serum T3, T4 and TSH. Administration of Hypericum Perforatum and memantine pre-scopolamine administration caused a decrease in the latency time in MWM and a decrease in the activity of ACHE and the contents of aspartic acid, glutamic acid, taurine, glycine and serine.
