Abstract This was performed by feeding rats with high fat (15%) and high fructose (20% w/v) diet for seven weeks (HF/FD). During the 6th week, all animals received a daily dose of dual-acting insulin (Mixtard 0.5 IU/Kg/i.p). At the beginning of 8th week, single subdiabetogenic dose of STZ (35 mg/kg/i.p) was given (HF/FD/STZ). Additionally, the present study was designed to investigate the effect of mangiferin (20 mg/kg/i.p), a xanthone glucoside from Mangifera indica bark, and valsartan (40 mg/kg/orally), an AT1R blocker, on insulin resistance, hepatic glucose output and metabolic disturbances of T2DM either alone or in combination with rosiglitazone (4 mg/kg/orally), a thiazolidinedione insulin sensitizer which is used as reference drug. Drug treatment with the various drugs started after one week of STZ injection and continued for 28 days. The effectiveness was tested after 14 and 28 days of treatment. |
