Abstract In the present study , eleven tumor samples (liver, spleen) were collected from layer and broiler breeder flocks in Egypt governorates in the period between 2009 to 2012. PCR assay was employed to detect Marek’s disease virus (MDV) in extracted DNA from tumor samples using specific primers target to 200 bp fragment of glycoprotein C (gC). Histopathological examination confirms MDV contribution. MDV were isolated using SPF ECE , SPF chicks and CEF. The isolated virus was molecularly characterized using PCR assay in extracted DNA from tumor samples and CAM of inoculated SPF chicks, and Sequence analysis confirmed the characterization of the MDV. Histopathological examination of organs of experimental infected SPF chicks and CAM and embryo of inoculated SPF ECE demonstrated the characteristic lesion of MDV. Propagation of MDV on CEF was applied for three passages, With no CPE was observed and PCR for extracted DNA of infected cells was negative for MDV. The present study reports the isolation and characterization of MDV associated with visceral tumors in layer and broiler breeder chicken flock in Egypt between 2009-2012. |
