Abstract This study aimed to : This thesis is devoted to the development of new methods for the analysis of some drugs used for the treatment of cardiovascular diseases in pharmaceutical preparations and biological fluids. The studied drugs are: aliskiren, hydrochlorothiazide, ramipril, metoprolol, felodipine and simvastatin. The thesis is devided into six parts: A general introduction about cardiovascular disease and classification of the drugs used for their treatment were described in the first part. A simple and specific method for simultaneous dtermination of aliskiren and hydrochlorothiazide in tablets and human urine by ion-pair liquid chromatography was developed. A simple and rapid HPLC derivatization method that enables sensitive determination of aliskiren in human plasma and in pharmaceutical formulations was developed. The simultaneous separation of 3 drugs (ramipril, metoprolol and felodipine) in a short run time with UV detection was achieved using a micellar liquid chromatographic method. The method was supported by appropriate experimental design and was applied for their analysis in tablets and spiked human plasma with acceptable limits of sensitivity. A simple stability-indicating micellar liquid chromatographic method with fluorescence detection was developed for the determination of felodipine in its tablets and spiked human plasma. The application of both high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC/MS) techniques for the determination of simvastatin in its tablet formulations as well as in spiked human urine was included in part six. The validation criteria of the developed methods were intensively studied. All the results were statistically analyzed and compared with those given by the official and comparison methods. |
