Abstract A grand total of 140 random samples of clams, mussel (50 of each) & oyster (40samples) were collected from Suez gulf and Alexandria governorate during summer of 2011 and 2012. All samples were subjected to bacteriological examination for detection of Vibrio species then confirmed by recent technique as PCR. The obtained results indicated that Vibrio species isolated from examined mussels samples collected from Alexandria and Suez gulf were 36% and 28 % respectively. The isolated serotypes were 1(4%) for each V. alginolyticus, V. furnissii and V. mimicus, 2 (8%) for each V. damsela, V. fluvialis , V. vulnificus for samples collected from Alexandria , and 1 (4%) for each V. fluvialis, V. hollisae and V. parahaemolyticus, 2( 8%) for both V. mimicus, V. vulnificus for samples from Seuz gulf. The multiplex PCR and rpoB sequence conformation for vibrio in mussel samples collected from Alexandria Governorate was 8% for V. cholerae and 4% for V. mimicus. But in Suez gulf was 12% for both V. cholerae and V. mimicus. The obtained results indicated that Vibrio species isolated from examined clam samples collected from Alexandria and Suez gulf were 44%and 24%, respectively. The isolated serotypes were 4 (20%) for V. alginolyticus,2 ( 8%) for both V. damsela and V. mimicus, 1 (4%) for both V. fluvialis and V. vulnificus for samples collected from Alexandria, and 2(8% )for V. alginolyticus, 1(4% )for V. hollisae, and 3(12%) for V. mimicus. for samples from Seuz gulf. The multiplex PCR and rpoB sequence conformation for vibrio in clams samples collected from Alexandria Governorate are negative. But in Suez gulf was was 4% for V. cholerae and 8% for V. mimicus. The obtained results indicated that Vibrio species isolated from examined oyster samples collected from Suez gulf was 45%. The isolated serotypes were 7(17.5%) for V. mimicus, 3(7.5%) for V. alginolyticus, 2(5%) for V. damsela, 2.5% for each V. fluvialis, V. furnissii, V. parahaemolyticus and V. vulnificus , 5% for mixed isolates. The incidence of pathogenic Vibrio species using multiplex PCR and rpoB sequence determination in Oysters samples collected from Seuz Gulf was 2.5% for both V. vulnificus and V. cholerae, 15% for V. mimicus, 7.5% for V. parahaemolyticus . from results of the current study, the combination of the multiplex PCR andrpoB sequencing allowed us to quickly and accurately identify the predominant Vibrio species that are pathogenic to humans,and not accuratly identified by biochemical methods due to relation ship between different species of vibrio as (V. vulnificus and V. cholerae ) and (V. cholerae and V.mimicus ) . These results indicated that Suez gulf samples were higher in Vibrio incidence when compared with Alexandria samples this may be due to the red sea higher temperature and salinity. The public health significance of the isolated Vibrio species (V. cholerae, V. mimicus, V. parahaemolyticus and V. vulnificus.) and probable occurrence in Egyptian shellfish were discussed. Furthermore, some recommendations to control such serious pathogens were mentioned. |
