Abstract

Cancer is clearly a worldwide problem. The incidence and mortality rates for various cancers are similar, though not identical, among developed countries. Cancer is a leading cause of death. Cancer is considered one of the major causes of mortality in the world. Despite the recent advances in science, cancer has not been cured yet. It is estimated that by 2020 there will be 16 million new cancer cases every year. The target of much research has been on the discovery of natural and synthetic compounds that can be used in the prevention and/or treatment of cancer. Natural products of either microorganisms e.g bacteria plant or animal origin that exhibited antitumor activity have been discovered. The main task of this study is to evaluate the antitumor activity of promising natural product compounds (azurin protein extracted from pseudomonas aeruginosa and Viscum album extracts) separately or in combination on cell lines in vitro and in vivo. In the present work, in vitro cytotoxicity screening of azurin protein from Pseudomonas aeruginosa and Viscum album extracted with 1-pure acetone,2-80%ethanol 3- boiling water and 4- cold distelled water ,and their combination on 4 differnent cell line cancer MCF-7, HeLa , EAC, and HepG2 by MTT assay method and estimating IC50 for all of them. Directly after 21 days of treatment, animals were anaesthised using diethyl ether, blood samples were collected from the heart by heparinized syringes for detection of the following parameters: SOD activity, CAT activity, GSH content, MDA concentration, ALT activity, AST activity, GGT activity,Urea concentration, and Creatinine concentration. The liver were dissected and kept in 10% formalin for histopathological examination.
Implantation of EAC into female Swiss albino mice has been proved to induce significant deleterious changes in antioxidant status. The results revealed a significant increase in lipid peroxidation level, a significant depletion in CAT activity, GSH content, and SOD activity in serum, of EAC bearing mice. Our biochemical findings were further supported by the histopathological examination of liver sections. The histological examination was as follows:- Group I:normal control mice shows normal hepatic architecture. Group II: bearing Ehrlich carcinoma showing focal aggregation of neoplastic cells. Group III: showed normalization of liver cells with regeneration of hepatocytes. Group IV: shows normalization of liver cells with regeneration of hepatocytes.
Group V: showing minute focal aggregation of neoplastic cells. Group VI: showing aggregated neoplastic cells. Group VII: showing generation of the hepatocytel s with minimal inflammatory infiltrate.
