Abstract Epitopes prediction plays a vital role in the development of antibodies production and immuno detection tests. The partial amino acid sequence was analyzed using DNAStar protean system in which four properties of the amino acid sequence were chosen as the parameters for epitope prediction including antigenicity, surface probability, hydrophobicity and secondary structure. These models were applied to potato virus y (PVY) as a case study to predict immunogenic peptides suitable for antibodies production. the most hydrophilic regions of PVY-CP were 36-40, 46-47, 67-74, 96-98, 130-133, 164-166 and 198-200 got high score and match the previous laboratory studies which make it one of the best candidates to be immunogenic and capable of producing antibodies that cross react with PVY. The peptide was chemically synthesized and injected into rabbit. The obtained antibodies were evaluated by using TAS-ELISA and immune dot-blot assay. The obtained antibodies were positively reacted against PVY infected potato tissues. Keywords: Immunoinformatics, ELISA, PAbs, potato, Potyvirus. |
