Abstract

Animals of 34 flocks contain 1257 native breed sheep found in Dakahlia Governorate were clinically examined from January 2015 to February 2016. Out of 1257 clinically examined animals , 43 animals were affected with clinical prevalence of ( 3.42% ). A cross sectional study was carried out between January and April 2016 to estimate the prevalence of CLA among sheep of Dakahlia governorate. The total number of sheep in the governorate was 1198500 according to the census of the governorate 2015. The total number of required animal samples was calculated using a simple random sampling design on Win Episcope 2. The total number of samples was 346 sheep.Out of 270 sheep, 171 were serologically positive with AP 63.33 % and 99 were negative .The true prevalence was estimated as 90.9 % and CI =95% ( 94.33-87.47 ). The highest locality prevalence was found among flocks of shirben (74.15%) then Mansoura ( 64.9% ) then Dekernes ( 58.9% ) while the lowest prevalence was recorded in Belkas ( 55.22% ). Out of 37 needle biopsy examined samples for PCR assay, 37 (100%) were positive rpoB indicated at 406 bp while 18 (48.64%) were positive PLD results indicated at 203 bp, from which 16 infected sheep and 2 incontact sheep but not detected in free sheep..
Sequencing of singleplex PCR products in order to confirm the mPCR results, ten (n= 2 PLD n=8 rpoB) randomly chosen PCR products were tested further .from 8 samples 4 samples from normal lymph node taken by needle biopsy showed that Klebsiella pneumonae and from 4 samples abscessiated 3 showed Stenotrophomonas maltophilia and one sample showed moraxella .
