Abstract The current study was performed to investigate age dependent immunoexpression and localization of somatotropin in the pituitary gland and some extra-pituitary organs. Twenty adult Wistar rats (fifteen females and five males) were housed; four animals per cage with observation of pregnancy. Samples from pituitary gland, liver, lung, testes and long bone were collected from different ages of male offsprings. Tissue specimens were processed and stained with H&E. Also, pituitary gland sections stained with special stain for detection of chromophilic cells. All tissue sections were stained with Avidin-biotin complex immunohistochemical technique to determine localization of somatotrophic hormone. Sections were divided into six stages according to the histological finding of pituitary gland. H&E results showed that there were histomorphological changes in pars distalis of pituitary gland at different stages. At stage I, acidophils were the distinguished cells; but basophils were difficult to seen then appeared at stage II. Immunoexpression of somatotropin in pars distalis showed that somatotrophs were numerous and uniformly distributed at first three stages; then they were restricted in clusters at last three stages. Somatotropin immunoreactivity of testicular sections showed a positive reaction in both the spermatogenic and the interstitial cells. it was detected in the lining epithelium of alveoli, interstitial tissue of pulmonary tissue and in cytoplasm of some hepatocytes; but it was absent in bone sections. |
