Abstract

This study was performed to improve the propagation of snowbush (Breynia disticha) family Phyllanthaceae by using two main methods of propagation technique (in vivo and in vitro propagation). Part one: the in vivo propagation was conducted to test the effect of cuttings types, auxin types (IAA, IBA and NAA) at various concentrations 0, 250, 500, 1000 or 2000 ppm. The best rooting percentage 91.7% was achieved with the terminal cutting treated with NAA at 500 ppm while, the best roots number 54 roots per cutting was achieved with terminal cutting treated with NAA at 2000 ppm. Part two: the in vitro propagation was included several stage, first stage surface sterilization with 3% sodium hypochlorite washing for 15 min was optimum for best survival percentage in shoot tips and nodal segment without contamination. Second stage was conducted to test the effect of two explant types (shoot tips and nodal segments) with four cytokinin types ( BA, Kin, TDZ or 2ip) at (0.1, 0.25, 0.5, 1.0 or 2.0 mg/L). The best response percentage (100%) was obtained with nodal segments with BA at 2 mg/L but the highest shoots number (3.83 shoots/explant) was obtained with nodal segments with BA at 0.5 mg/L. After there, second experiment was conducted to test the effect of two explant types, auxins (IBA, NAA or IAA) at (0.05, 0.1, 0.25 or 0.5 mg/L) with BA at 0.5 mg/L (the best result of previous experiment). The best response percentage and number of shoots (100 % and 3.56 shoots/explant) with IBA at 0.5 mg/L + BA at 0.5 mg/L. Third experiment was conducted to test the effect of two explant types and various concentration of GA3 (0.05, 0.1, 0.25 and 0.5 mg/L) with BA at 0.5 mg/L. The best result was recorded at 0.1 of GA3 + BA at 0.5 mg/L gave the shoots number (5.25 shoots) and success to enhance rooting percentage of 50 %, number of roots and root length of 3.25 roots per shoot and 5.35 cm, respectively. Rooting stage the best number of roots (9.5 roots/shoot) was recorded with NAA at 2 mg/L and active charcoal while, length of root (7.33 cm) was recorded with IBA at 2 mg/L and active charcoal.
