Abstract Diabetes is considered a life threating disease due to its complications. Type 2 diabetes accounts for about 90% of wholly diabetic cases. There is a need to find new strategies to achieve the management of type 2 diabetes to avoid its complications and side effects of diabetic drugs. The aim of this study was to investigate the anti-diabetic effect of Curcumin-Magnesium oxide nanoparticles conjugate in type 2 diabetic rats. This study included 5 groups: group 1 (control group), group 2 (type 2 diabetic rats), group 3 (type 2 diabetic rats were received Cur-MgO NPs conjugate (5mg/Kg/day for 45day, orally)), group 4 (type 2 diabetic rats were received Cur-MgO NPs conjugate (10 mg/Kg/day for 45day, orally)) and group 5 (type 2 diabetic rats were received Cur-MgO NPs conjugate (20 mg/Kg/day for 45day, orally)). The levels of plasma glucose, antioxidant enzymes (SOD and CAT ), lipid profile, liver enzymes (ALT,AST and ALP) and MDA were measured. Moreover, histopathological examination of liver and spleen was examined and gene expression of ACC, FAS, PPAR alpha and MCD. The levels of glucose, TC, TG, LDL-C, VLDL-C, ALT, AST, ALP and MDA were normalized in the treated groups in compared with diabetic group. While level of SOD, CAT and HDL-C were not significantly changed between all groups. In the histopathological examination of liver showed severe hydropic degeneration with individual coagulative necrosis of hepatocytes which improved gradually with increasing the dose in the treated groups. Spleen showed damage of lymphoid cells in T2D while in treated group showed improvement in spleen tissue and cells are rescued from apoptosis gradually with increasing the dose. In Gene expression, the level of mRNA ACC and FAS were significantly increased in diabetic group then normalized to the control group in the treated groups. While, mRNA level of PPARα and MCD were significantly reduced in the diabetic group then normalized to the control group in the treated groups. The data of this study revealed that Cur-MgO NP conjugate has potential ameliorative effect on blood glucose, lipid profile parameters, liver enzymes, MDA, liver and splenic tissue as well as level of (ACC, FAS, PPAR alpha and MCD) genes in type 2 diabetic rats. |
