Abstract Methotrexate is a common chemotherapeutic agent, a structural analogue of folic acid, MTX can cause acute, subacute and long-term neurotoxicities. The mechanism of neurotoxicity is likely through disruption of CNS folate homeostasis and/or direct neuronal damage by oxidative stress and lipid peroxidation. Lepidium sativum seeds had a marked influence on fracture healing in rabbits, clearly supporting their effects on human beings, as a well –known natural element to promote fracture healing in traditional medicine. Many studies on seeds of L. sativum have suggested the antioxidant effect of lepidium. The aqueous extract of L. sativum seeds has been reported to have anti-hypertensive and diuretic effect when studied in normotensive and spontaneously hypertensive rats. The effectiveness of this plant in treatment of bronchial asthma, hiccups, cough with expectoration. Oxidative damage and the formation of free radicals may occur for several reasons such as exposure to chemicals, metals, irradiation and toxins causing to lipid peroxidation, which in turn affects the activities of protective enzymatic antioxidants that are greatly sensitive indicators of increased oxidation reaction. The present study was aiming to study the effect of methotrexate on cerebellum of adult male albino rats and the possible protective role of Lepidium sativum based on histological and immunohistochemical study.
