Abstract This thesis is devoted to design, synthesis, biological evaluation, enzyme activity inhibition assay, caspase-3 assay and apoptosis induction, cell-cycle analysis and molecular modeling of novel benzofuran derivatives as antitumor agents based on molecular hybridization strategy. Part I is a general introduction about cancer, anticancer drugs and the studied drugs including benzofuran derivatives as antitumor agents. Part II is a research objective which is design and synthesis novel benzofuran derivatives as antitumor agents based on molecular hybridization strategy. Part III represents theoretical discussion of design thirty four new compound. Fourteen derivatives had strong antitumor activity, three derivatives has Top I inhibition activity and six derivatives has EGFR inhibition activity. Part IV represents experimental works for design thirty-four compounds which synthesized for the first time. Reaction times were monitored using TLC plates, the target compounds were purified by silica gel column chromatography or by recrystallization and the structure elucidation of these compounds were fulfilled based on spectra analysis, IR, 1H-NMR, 13C-NMR, mass spectroscopy and elemental analyses. This thesis consists of four parts including 10 tables, 43 figures, 43 spectrum and 233 references and ends with an Arabic summary. |
