Abstract The present study was aimed to study the effect of Olea europaea leaves extract and Silybum Marianum seeds extract on diabetic rats induced by STZ, and carried out the following investigations to fulfill the following objectives :1- Explore the in vitro antioxidant activity of Olea europaea leaves extract and Silybum Marianum seeds extract.2- Investigate the hypoglycemic effect of Olea europaea leaves extract and Silybum Marianum seeds extract and their combination in Streptozotocin induced diabetic rats.3- Evaluate the effect Olea europaea leaves extract and Silybum Marianum seeds extract and their combination on oxidative stress associated with diabetes mellitus.4- Study the molecular mechanisms through important biological parameters including GCK, apoptotic markers (Bcl-2 and Bax) and antioxidant status (Nrf-2 and HO-1) for their antidiabetic activity.5- Study histopathological changes of pancreatic tissues associated with diabetes mellitus after the treatment. from the present study, it may be concluded that the treatment of diabetic rats with extracts of OL or SM or their combination (250 mg /kg body weight /day) showed a marked improvement in the effects of STZ, which led to a decrease in the level of blood glucose and glycosylated hemoglobin and increase insulin levels, as well as reducing oxidative stress through stimulation the activity of SOD and CAT. It also inhibited the production of MDA. On the other hand, the treatment of diabetic rats with these extracts resulted in elevation of gene expression of pancreatic GCK and Bcl-2 genes and suppressed the expression of Bax gene. However, there are up-regulation of hepatic Nrf-2 and HO-1 genes. In addition, the pancreatic cells were restored to produce the insulin. |
