Abstract

The thesis is divided into five main parts :- Part I. includes general introduction about investigation and development of recently approved drugs. Part II.A. includes an introduction to quantitative proton nuclear magnetic resonance technique. Part II.B. includes introduction to betrixaban, fluticasone, azelastine and their reviewed methods for determination of these drugs in raw materials and pharmaceutical dosage forms. Part II.C. includes a quantitative proton nuclear magnetic resonance method for quantification of a novel anti-coagulant drug (betrixaban) in its pure form and laboratory prepared dosage form. Moreover, the study included the expected degradation pathway of betrixaban which was derived by assessing the stability of the drug under different stress conditions according to ICH guidelines. Part II.D. includes a quantitative proton nuclear magnetic resonance method for simultaneous determination of newly approved combination of fluticasone and azelastine in bulk and pharmaceutical preparation. Part III.A. includes introduction to capillary electrophoresis technique. Part III.B. includes an introduction to budesonide and reviewed methods for its quantification in raw materials and pharmaceutical dosage forms. Part III.C. includes a micro-emulsion electro-kinetic chromatography method coupled with field amplified sample stacking for simultaneous determination of novel combination of azelastine and budesonide in their pure form and combined pharmaceutical preparations. Part III.D. includes a comparative analytical study between different modes in capillary electrophoresis for quantitative determination of betrixaban in pharmaceutical and biological matrices. Part IV. includes HPLC method for determination of betrixaban with a co-administered drug lercanidipine in their laboratory prepared mixture, laboratory prepared dosage form as well as in spiked human plasma. Part V. includes four spectrophotometric methods for determination of two different anti-thrombotic drugs, the anticoagulant; betrixaban and the antiplatelet; aspirin, alone and in their novel combinations.
