Abstract The aim of the present study was to evaluate the value of intra-peritoneal administration of CBL through comparasion the different duration and assessment of functional recovery of diaphragmatic muscle by using Electromyography (EMG) and histopathological evaluation of spinal cord in Sprague-dawely rats that undergo cervical spinal cord hemisection. The present study was carried out on a total number of 32 female Sprague–Dawley rats (body weight 200–250 gram). Animals were divided into 3 main groups and the third group is divided into two subgroups. group I: Negative control; normal rats without cervical spinal cord hemi section. group II: spinal cord injury untreated; all rats subjected to cervical spinal cord hemi section without treatment (positive control).group III: spinal cord injury treated with cerebrolysin Subgroup III a) in this group, cervical spinal cord hemi section was performed and each animal received 2.5ml /kg cerebrolysin IP for 7 successive days. Subgroup III b) in this group, cervical spinal cord hemi section was performed and each animal received 2.5ml /kg cerebrolysin IP for 21 successive days. Functional assessment of diaphragmatic ms paralysis was monitored after 28 days post-surgery using Biopic system Inc. with electrode MEC110C. All rats in each group were euthanized using urethane 97% in a dose of 1.5g/kg I/P at 28th days post operation. Histological Analysis at the end of the study for all rats for H&E, Cresyl Violet stains and in GFAP immunoperoxidase was performed. H&E stain and cresyl violet stain were used to characterize the spinal tissue architecture in all groups. Additionally to that, the GFAP immunoperoxidase was applied to examine the distribution and activity of astrocytes. The results of the present study demonstrated that the rats which received cerebrolysin for successive 21 days post injury showed significant increase in left diaphragmatic ms activity compared to positive group and 7 days treated group. Meanwhile the group which received cerebrolysin for 7 days post-surgery showed slight significant difference compared with control positive group. |
