Abstract

The purpose of this study was clarify the immunomodulatory effect of Echincea Purpurea and Orguinum Vulgaris via evaluation of different immune biochemical parameters in serum , MTT assay for lymphocytes proflieration , histological examination and immunohistochemistry of the spleen of albino rat. The study was designed as follow: eighty mature rats were divided into four equal groups, (G1) non-treated, (G2) that orally administrated 300mg /day of Echinacea extract (EP) in drinking water., (G3) rats which administrated 200mg /day of (OV) in drinking water, and (G4) rats that administrated mix of (EP) 150mg /day and (OV) 100mg /day for two weeks. Blood samples of rats were collected at 1,3,5,7,9,12,15 days following the first day of administration of herbal extract in drinking water through median cancuc vein of eye there for investigation of the effect of (EP) and (OV) extracts each alone or mix of them on serum biochemical parameters including total protein, albumin and globulin. After two weeks, rats of the four groups were sacrificed and spleen tissues of rat of each group were collected and fixed then processed for histopathological examination and stained with hematoxylin and eosin and immunohistochemistry for quantitative evaluation of CD3, CD20 and Caspase-3 as well as histomorphometry analysis.
Results: MTT lymphocyte proliferation assay revealed marked increase from day one compared to control, this increase was moderate in day five and in day fifteen showed mild elevation G4 then G2. Histopathological examination of spleen tissues compared with (G1), rat spleen of (G2) revealed moderate hyperplasia of white pulp. On the other hand, (G3) showed moderate hyperplasia of red pulp with accumulation of megakaryocytes in large number. Noticed mild, herbal cytotoxic changes in red pulp and splenocytes of both G2 & G3. Those cytotoxic changes were not noticed in G4. Immunohistochemically, quantitative evaluation of CD3+ T-lymphocyte intensity and distribution, in G2, G3, G4 comparing to the control G1 which is (47%), showed 50%, 53%, and 62% increase respectively.
Conclusion: this study verified that EP and OV extract and mix both of them had improvement effect on rats ,the mix of lower doses of the two immunomudators in group 4 give more improvement to all parameters were measured ,we suggesting that synergistic enhancing effect was done between the two modulators .
