Abstract

In the chicken industry Mycoplasma and Salmonella are vertically transmitted diseases that cause significant economic losses, so this study aimed to prepare locally an inactivated bivalent Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) vaccine and pentavalent Salmonella Typhimurium (S.T.), Salmonella Entriditis (S.E.), Salmonella Kentucky (S.K.), (MG) and (MS) vaccine also, trivalent S.T., S.E. and S.K. commercially available vaccine was evaluated. The prepared bivalent and pentavalent vaccines were adjuvanted with Montanide ISA70 then tested for safety, sterility, and potency. Both vaccines efficacy was evaluated in 160 SPF one day old chicks. Chicks were vaccinated with 1st dose at a week of age then boostered with the 2nd dose after 3 weeks. Three weeks post vaccination with 2nd dose chicks were challenged, the birds were kept under observation for 10 weeks. During the experiment clinical symptoms, daily mortalities, postmortem examination, and weights were recorded as well as, serum, cloacal swabs and nasal swabs were collected. Mycoplasma and Salmonella strains humoral immune response was assessed using serum samples taken weekly after first immunization until the third week after challenge measured by the enzyme-linked immunosorbent assay (ELISA) test and results showed that vaccines gave a high protective antibody titer. We concluded that locally manufactured inactivated bivalent Mycoplasma vaccine gave 85% protection, trivalent Salmonella commercially available vaccine gave 80% protection and pentavalent vaccine gave 100% protection. Based on our results, we recommend using of a locally prepared inactivated pentavalent vaccine to control of mycoplasmosis and salmonellosis in layers and breeders chicken farms in Egypt.
