Abstract The present study was designed to investigate the effect of addition of different concentration of cysteine (0 ,0.6 0.8 1 Mm) into maturation medium on cumulus expansion , the ultra-suture of the oocytes ,the expression of BMP15, GDF9 and CB-1 genes in bovine oocytes, the developmental competence of bovine oocytes. Cow’s ovaries were collected within 0.5 h after slaughtering and transferred to Lab.in sterile saline with the antibiotic in a thermos container. Cumulus oocytes complexes (COCs) were aspirated from all antral follicles. Oocytes with homogenous cytoplasm and surrounded with 2 or more layers of compact cumulus were matured in-vitro in 4-well plastic petri dishes for 24 hrs with different cysteine concentration (0 ,0.6, 0.8 ,1 mM) at 38.5 ºC and under 5% CO2. In all experiments, some of the in-vitro matured oocytes were used as COCs and the others were denuded through repeat pipetting. In first experiment COCs matured with different concentrations of cysteine the percentage of expanded COCs as well as the degree of its expansion was detected after maturation. A significantly higher values were recorded in all L-cysteine treated groups compared to control group, with the best expansion degree for 0.8 mM L-cysteine group. |
