Abstract 56 samples of blue crab Callinectes sapidus obtained seasonally from Mediterranean coast of Port Said city during 2020 year. This study was planned to define seasonal biometric relationship in relation condition factor, and to determine the proximate composition, minerals, lipid classes, antioxidant and immune enzyme activities, heavy metals, organochlorine pesticide residues, and isolation and identification of bacterial species from hemolymph. the samples were weighed to nearest gram, and carapace width and length (mm) were measured by vernier caliper. Hemolymph was extracted by sterile syringe seasonally. The samples were dissected to obtain hepatopancreas, gill, and muscle tissue, the association between carapace width and carapace length was expressed using the linear regression analysis CW=b CL+ a: CW=carapace width, a=Intercept, b=regression coefficient(slop),CL=carapace length, and the length-width/weight relationships were calculated using the formulas W=a CL b and W=a CW b, where W represents the weight (g), CL represents the carapace length (mm), CW represents the carapace width (mm), a represents the regression intercept, and b represents the regression coefficient, Dubios et al. 1956 technique was used to determine the total carbohydrate content. The folin-ciocalteu phenol technique was used to determine total proteins in muscle tissue samples. Lipid was extracted using the chloroform method of extraction. Calcium, phosphorus, sodium, potassium, and sulfur in muscle tissue, measured in mg/g in a spectrophotometer using a colorimetric approach. Cholesterol, triglycerides, and phospholipids in muscle, hepatopancreas, and gill samples were measured using –enzymatic colorimetric technique. The antioxidant and immune enzymes activity were determined by using colorimetric technique. Heavy metal levels were measured using a Buck scientific Accusys 211 Atomic Absorption spectrophotometer USA.The OCPs residues in hepatopancreas, gills, and muscles were obtained by using Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) method, The bacteria species were identified using the Micro -SCAN-10. The total weigh/ carapace width-length,and carapace width/length ratio were negative allometric in both males and females, body weight was higher in male than female and condition factor of females were higher than males in all seasonsThe nutritional constituent of males were higher than that in females. The highest concentration of heavy metals were in summer and the lowest noted in winter, the concentration of heavy metals were below the permissible limit recorded by FAO and USDA(2003).The only organochlorine pestcides residue detected was o’p-DDE, and it’s level was below permissible limit recorded by FAO and USDA(2003).The bacterial species identified seasonally in Hemolymph were Klebsiella, Achromobacter, Micrococcus, Kocuriarhizophilia, Staphylococcus, Yersinia enterocolitica, and Seratia. |
