Abstract

The Musca domestica salivary gland hypertrophy virus (MdSGHV) is a double-stranded DNA virus that replicates in the salivary glands of infected adult house flies that inhibits egg production in infected females and is characterized by salivary gland hypertrophy (SGH) symptoms. In this study house flies infected with (MdSGHV) were found in fly populations collected from Giza and Qalubia governorates, Egypt livestock locations that were surveyed in 2010 and 2012. Transmission of this virus is believed to be mediated orally via deposition and consumption of oral secretions composed of salivary gland secretions and crop contents transmission electron micrographs of crops from infected flies showed numerous enveloped virions, as well as on the salivary gland cells. Oral treatments of newly emerged flies with viremic salivary gland homogenates, virus resulted in an average 20-55% infection. Crude extract from SGH resulted in a positive PCR reaction equivalent purified DNA was used as the template. In conclusion, PCR using a purified DNA template from an infected SGH, method for virus diagnosis in live house flies. Isolates were examined by using molecular approaches, 5 genes (p74, pif1, pif2, pif3, DNA polymerase) were partially sequenced to determine and distinguish the MdSGHV isolate
