Abstract This work was carried out mainly in the Tissue Culture laboratory of the Plant Production Department, the Faculty of Agriculture, Saba Basha, Alexandria University, and the Plant Molecular Biology Lab., & Physiology Plant Tissue Culture Lab., Department of Plant Biotechnology, Genetic Engineering and Biotechnology Research Institute (GEBRI), University of Sadat City, Egypt, during the period of 2016- 2019. In order to efficiently maximizing the induction of plants via direct organogenesis, it was important to study explant surface disinfectant as well as the influence of different growth regulators alone or in combinations along with the tissue culture stages and germination and finally morphological assessing of the regenerated plants from in vitro culture and comparing them with their mother stock plant at the molecular level. Therefore, the experiments of the present work were divided into two parts. The first part is micropropagation was included sex experiments: 1. The Effect of type and concentrations of different disinfectant on surface sterilization. 2. Effect of growth regulators on type of explant on starting stage (Internode cuttings - Seeds). 3. Effect of type and salt strength of (WPM – MS) culture medium. 4. Effect of growth regulators on Multiplication stage. 5. Effect of growth regulators on rooting stage. 6. Effects of mixture type on vitroplants in acclimatization stage. The second part Assessment of genetic stability of produced vitroplants: 1-1 Random Amplified Polymorphic DNA (RAPD). 1-2 Inter-Simple Sequence Repeat (ISSRs). |
