Abstract This part involves a general introduction about the studied drugs and their related impurities and covers their pharmacological action, chemical structure, physical and chemical properties, and literature review.Part II: HPLC determination of Xipamide, Triamterene, and toxic impurity; Benzyl cyanide with greenness assessment. In this part, HPLC method was developed for simultaneous determination of a mixture of antihypertensive drugs, Xipamide, Triamterene, and toxic impurity; Benzyl cyanide in pure form and pharmaceutical formulation. Optimum chromatographic separation was achieved with the required symmetry, accuracy, and precision using Shimadzu HPLC C8 column (150 × 4.6 mm ×3 μm particle size) with a mobile phase consisting of 0.05 M KH2PO4 (adjusted to pH= 4.5 by phosphoric acid): methanol (50: 50 v/v) of flow rate 1.2 mL/min and the eluate was scanned at 254 nm at temperature (30ºC). Calibration curves were constructed in the range of 1 – 100 μg/mL for both Xipamide and Triamterene and 50– 50 μg/mL for Benzyl cyanide. The greenness of the proposed method was assessed using the Analytical Eco-scale, and the Analytical Greenness calculator (AGREE) metric tools and compared with the reported method and achieved high scores. |
