Abstract In the present study a total of 300 samples from apparently healthy and diseased living and slaughtered cattle were examined for detection of Mycoplasma spp., after primary isolation found that 137 positive isolates of total 300 examined samples with an incidence 45.6%, apparently healthy living cattle 20/46 (43.4%), diseased living ones 64/89(71.9%), while apparently healthy and diseased slaughtered cattle were 25/131(19.1%) and 28/34(82.3%) respectively. Degitonin sensitivity test showed that 90 Mycoplama isolates from total 137 positive examined samples. Biochemical Identification of the isolated Mycoplasma revealed more than one species of Mycoplasma; the most important one was M.bovis in percent of 32.2% from the total isolates, then M.arginini in 22.2% and M.bovirhinis in 20%. Application of PCR for confirmation of Mycoplasma isolates by detection of 16S rRNA gene and using specific primers for M.bovis, M.arginini, M.bovirhinis and detection of vsp gene of M.bovis followed by Nucleotide sequence analysis of Mycoplasma spp. and vsp gene |
