Abstract This study was conducted to to evaluate retrospectively the effects of genetic polymorphisms of CYP3A4, CYP3A5 and MDR1 on cyclosporine trough and 2 hour s blood concentration during the early period after renal transplantation in Egyptian patients. Forty two renal transplant recipients and forty eight healthy controls Egyptian subjects were genotyped by polymerase chain reaction{u2013}restriction fragment length polymorphism for CYP3A4*18A, CYP3A5*3 and MDR1 C3435T. Cyclosporine C₀ and C₂ whole blood levels were measured. The frequency of the CYP3A4*18A, CYP3A5*3 and MDR1 C3435T variant alleles in all subjects were 0.006, 0.865 and 0.344 , respectively,comparison of the CsA levels between patients with CYP3A5 3*/3* genotype and those with 1*/1* genotype, showed that all levels of C₀ and C₂ are higher in 3*/3* genotype than that of 1*/1* genotype but without significant differences between the two groups at day-7 C₀ (P = 0.25), day-30 C₀ (p = 0.28), day-90 C₀ (p = 0.3) and at day-30 C₂ (P = 0.64) |
