Abstract Diclofenac is a non-steroidal anti-inflammatory drug. It is generally used for the treatment of pain, arthritis and gout, not only in the humans but also in the animals. It inhibits the synthesis of prostaglandins from the archidonic acid by inhibiting the cyclo-oxygenouse 2 activity. Diclofenac sodium has adverse effects on the hematological parameters, gasterointestinal tract, hepato-renal structure and function, spleenic structure, myocardial structure and brain structure. The diclofenac sodium is safe at therapeutic dose, but it is toxic when used at overdoses. The toxic effect of DS may attributed to the oxidative stress induced by this drug, consequently upon leading to the generation of free radicals which disturp the structure and function of the biological system. Silymarin is a polyphenolic flavonoids extracted from fruits and seeds of milk thistle. Silymarin acts as antioxidant, free radical savenger, cell membrane stabilizer and permeability regulator. Due to its antioxidant properties, silymarin is a well tolerated and effective antidot for the use in the protection of the biological system against the toxicity of many harmfull material and toxic substances. However, its mechanism of action is still poorly understood. |
