Abstract The present work is the first to evaluate the chronic effects of AFB2 in comparison to aflatoxin mixture on histochemical and immunilogical parameters in albino rats. 60 rats ( 70+1.5 g) were divided into 3 equal groups, the first was fed on the basal diet and kept as control ,the second was fed on a diet containing 1.0 ppm AFB2 and the third was fed on a diet containing 1.0ppm of aflatoxin mixture too all rats were fed for 6 weeks as treatment period followed by 2 weeks withdrawal period. After 2 and 6 weeks of treatment rats were sacrificed to investigate the examined parameters.The results can summarized as following : Both treatments showed a reduction in growth performance , the plasma concentrations of glucose , cholesterol , total protein and albumin . In similar, the activity of plasma ALT, AST, ALP and ACP. The withdrawal period, most disturbances of biochemical parameters were partially recovered in both treatments. Also, haematological and immunological parameters both treatment caused asignificant increases in RBCs, Hb content, WBCs And more potent in rats fed on AFB2 than those fed on the mixture. Histolopathological and histochemical altrations were observed in the studied sections under the influence of AFB2 and aflatoxin mixture, lymphocytic infiltration, pyknotic nuclei, vacuolar degenerations, haemorrhage and increased Kupffer cells were recognized in aflatoxin treated liver and kidney. In treated testes, distruction of the seminiferous tubules, haemorrhage, vacuolations and delay in the process of spermiogenesis were observed. Marked decrease in PAS positive materials in the examined organs, also, the same results seemed to appear in the tissues stained by PAS Alcian blue to demonstrate the mucopolysaccharides. Total protein stained by bromophenol blue which appear as great loss of protein stained materials in both treatment but AFB2 was more effective. Nucleic acids were observed in the examined sections under the influence of both treatment by using Feulgen and Pyronin u, the toxins displaying marked decrease in nucleic acids. AFB2 was more effective. The withdrawal period could partially recover and nearly restore the normal configuration of the studied organs in all histological and histochemical demonstrations. Furthermore, this study represents an interesting point for further multidirectional studies on the action and metabolic pathways of aFB2 |
