Abstract Isolation of some bacterial isolates and screening of them using Nile Red stain test. Optimization for the 4 promising isolates in different growth media with different growth condition. Extraction of the biopolymer and analysis for this biopolymer using IR and HNMR, and the analysis improved that the resultant biopolymer is polyhydroxybutrate (PHB).To increase the productivity of the isolate S4 physical and chemical mutations were carried out. To obtain transgenic bacteria accumulate a high amount of the PHB inside their cells. Also optimization for the transgenic bacteria was done on non expensive media. Semiindustrial production of biopolymer was carried out by growing the transgenic bacteria on 3 litter fermentor. Identification of the wild strain using 16SrRNA gene technique and also the synthase gene was identified. |
