Abstract

Diabetes mellitus type 2 is the most common form of diabetes (90–95%) and is due to insulin resistance or reduced insulin sensitivity, combined with reduced insulin secretion. The aim of the present study was, first to induce animal model of insulin resistance by feeding rats on high sucrose diet. Secondly, to evaluate the beneficial versus harmful effects of thiazolidinediones(rosiglitazone and pioglitazone) on both vascular system and liver The data obtained in the present study showed the following: 1. Administraiton of high sucrose diet to rats for 8 weeks resulted in significant increase in body weight,plasma triglyceride, plasma glucose, plasma insulin and plasma TNFα together with insignificanct effect on plasma total cholesterol level. Also, high sucrose diet to rats produced significant disturbance in the vascular outflow in perfused hindlimb, significant intimal proliferation in coronaries and marked hepatocellular degeneration, necrosis and infiltration of inflammatory cells, and fibrosis with multiple foci of micro- and macro-vesicular fat accumulation, hepatic lipidosis in the liver tissues. 2. Administration of rosiglitazone for 6 weeks produced significant increase in body weight together with significant decrease in plasma level of triglyceride , plasma glucose, plasma insulin, plasma TNFα and insignificant effect on plasma total cholesterol level. Also, treatment with rosiglitazone produced significant improvement in the vascular ouflow rate in perfused hindlimb, significant reduction in intimal proliferation in coronaries and milder hepatocellular degeneration, necrosis and infiltration of inflammatory cells, and fibrosis but with greater multiple foci of micro- and macro-vesicular fat accumulation, hepatic lipidosis in the liver tissues 3. Administration of pioglitazone for 6 weeks produced significant decrease in plasma triglyceride, plasma glucose, plasma insulin and plasma TNFα together with insignificanct effect on body weight and plasma total cholesterol level. Also, produced significant improvement in the vascular ouflow rate in perfused hindlimb, significant reduction in intimal proliferation in coronaries and milder hepatocellular degeneration, necrosis and infiltration of inflammatory cells, and fibrosis with less multiple foci of micro- and macro-vesicular fat accumulation, hepatic lipidosis in the liver tissues.
