Abstract 1 Make growth curves and some biochemical properties (Thermal stability; pH activity; kinetic determinations and molecular weight of the purified enzyme) of rawstarchdegrading gasamylases produced from different yeasts. 2 Isolation of gasamylase gene and introduced into industrial yeast strains using CaCl2 and PEG methods. 3 S. cerevisiae transformants harboring gasamylase gene were tested for their ability to utilize starch and secretion of gasamylase activity on liquid and solid soluble starch media. 4 Determination of genetic stability to different recombinant yeasts. 5 SDS polyacrylamide gel electrophoresis (SDSPAGE) of S. cerevisiae transformants and amylolytic yeast strains: a Molecular weight of protein bands in amylolytic strains and their transformants. bThe similarity and genetic distance between amylolytic yeast strains and their transformants based on protein patterns. 6 Evaluation S. cerevisiae transformants to secretion of amylolytic activity and ethanol production on potato starchy wastes. In order to uses this transformants from S. cerevisiae in assimilation industrial wastes which containing starch and its bioconverted to ethanol and biosafety single cell protein, so, this study may be very important in application of recycling environmental wastes containing starch by using recombinant S. cerevisiae strains. |
