Abstract

Toluene is widely used in industry, and is applied mainly as a solvent in paints, lacquers, and adhesives. Toluene has adverse effects on male reproductive functions; testicular atrophy and reduced spermatogenesis. Therefore, the aim of this work is to demonstrate the relation between age and testicular toxicity caused by toluene in male rats after one month of toluene exposure.. Male Wistar rats of age 42 days old (28-33 g), 52 days old (45-50g) and 70 days old (75-80 g) representing (prepuberty, puberty and sexually mature ) age stages were studied. They were divided into three main groups: At the end of treatment period, the animals were scarified and the serum, testis were collected to determine some physiological parameters such as level of testosterone hormone, some enzymes 3β-HSD, G-6-PD, LDH, ALP,SOD lipid perioxidation products and total lipids, triglycerides and phospholipids. Histological studies as well as morphometeric analysis to the testis were also carried out. The following changes were recorded in toluene groups comparing to normal control as well as olive oil groups: 1-A decrease in serum testosterone level in all treated animals, age 42 days was the more affected age. 2-A decrease in the activities if 3β-HSD,G-6-PD, LDH and ALP in testis. 3-A decrease in activity of antioxidant enzyme SOD accompanied by an increase in lipid peroxidation product and in testis, age 42 days was the more affected age. 4-A decrease in total serum and testis proteins, age 42 days was the more affected age. 5-A decrease in DNA and RNA content in testis. 6- An increase serum total lipids, triglycerides whoever, a decrease in serum phospholipids levels was recorded. 7-A decrease in testis total lipids, triglycerides and phospholipids. The histological observations revealed that toluene exerts histological effects on testis including; vacoulation, necrotic cells with nuclear pyknosis while testis of control and olive oil groups showed normal structure.
