Abstract This study aimed to evaluate the antitumor effect of silymarin against DEN/CCl4-induced HCC in rats biochemically and histopathologically. eight adult male albino rats weighed (100-120 gm) were used in this study and were divided to 4 groups. In the first group (control group), in second group rats were supplemented with 1000mg/kg silymarin. In group three, four HCC model induced by single I.P. injection of DEN with a dose of 200 mg/kg followed by subcutaneous injection of CCl4 (3 ml/kg) weekly. In group four supplemented with silymarin simultaneously with cancer model. result of this study showed that the activities of ALT, ALP and level of total bilirubin and direct bilirubin in serum were increased in group three, while they were decreased in group four. The level of albumin in serum was decreased in group three, while it was increased in group four. The levels of oxidative stress marker as (MDA and H2O2) in liver tissue were increased in group three, while they were decreased in group four. The antioxidant status in liver tissue, which was evaluated by measuring activities of SOD, catalase and level of GSH, were decreased in group three and improved in group four. The expression of p53 was down-regulated in group three, while it was upregulated in group four. However, the expression of MMP-9 was upregulated group three, while it was down-regulated in group four. The histopathological examination showed the lesions of HCC in group three, which was alleviated in group four. The expression of CD 10 by immunohistochemistry and expression of Ki67 by flow cytometry were increased in group three, while they were decreased in group four. |
