Abstract

This study aimed to determine the effect of to evaluate the effect of addition of vitamin C (1 mg/ml of vitrification solution) or zinc chloride (1.5μg/mL of vitrification solution) and Nano- zinc oxide (1 μg/mL of vitrification solution) to vitrification medium on morphological viability, staining viability, ultrastructural changes and in vitro maturation of vitrified immature (local breed) bovine oocytes. Evaluate. Compact cumulus oocytes (n=2022) were obtained from slaughtered bovine ovaries. Then the morphology of oocytes was examined using a stereomicroscope. Staining with trypan, the in vitro maturation, and Electron microscopy, after vitrified –warming and maturation the oocytes ultrastructural changes were studied. The results revealed the effect of vitrification media on total survival rate of oocytes relative to vitrified oocytes was affected significantly, ranging between 90.28% with ZnCl in vitrification medium, with Nano-ZnO 87.22%, with Vit.C 82.5% and with vitrification medium alone 74.44%. Relative to total number of vitrified oocytes, post-thaw survival rate of oocytes in normal type was significantly (P<0.01) higher in case of using the vitrification medium with ZnCl and Nano-ZnO than that with Vit.C and control (81.11 and 79.44% vs. 65.56 and 54.74%, respectively). Meanwhile, survival rate of oocytes in abnormal type was significantly (P<0.000) the lowest (7.78 and 9.17%) in vitrification medium with Nano-ZnO and ZnCl were higher (16.94%) in vitrification medium with Vit.C. and (19.72%) in control. The ultrastructural alterations in perivittaline space and cortical granulosa were showed highest in bovine oocytes vitrified control followed by ZnCl group then Nano ZnO group. Whoever, the ultrastructural alterations in mitoconderia was higher in vitrified oocytes without supplementation than other groups. Conclusion, supplementation of vitamin C or Zinc chloride to the vitrification medium improved, morphologically and ultrastructural survival and viability rates as well as maturation rate of balidi cows oocytes. But the best results were the addition of Nano ZnO to vetrification medium.
